Does a nonprofit employer have to show that they couldn't find US citizens/ residents with comparable qualifications for the job, before sponsoring hiring a worker who will require H1B sponsorship?

The rules are the same for every H1 employer.

So does any employer have to show that there are not enough US citizens/ residents for hire for the given job while petitioning for the H1B?

For an H1, that doesn't have to be shown. For a GC, that has to be shown.
